ES細胞を用いたキメラマウス作製法

The method of producing the chimeric mouse with embryonic stem cells
Summary The mouse that modifies the mouse chromosome freely according to the purpose can be made as a method of producing the mouse that modifies the gene in the gene targeting mouse producing system that use embryonic stem cells (ES cells) though there are a method of injecting DNA directly into the fertilized egg and using the virus. This method was established to the technology that made the chimeric mouse by using ES cells that had been established in the 1970's by combining with the technology that did the homologous recombination to ES cells. ES cells cultured with the early embryo differentiates under normal embryo generation, and forms the chimeric mouse composed of the host embryo cell. Because ES cells differentiates into chimeric mouse's germ-line cells, the gene mutation of ES cells is inherited to the descendant mouse since the next generation by mating of the chimeric mouse and the wild-type mouse, and a new strain of mouse is created.
